TMB (tumour mutational burden)
Other names: tumor mutational burden, beban mutasi tumor
TMB measures how many changes are present in tumour DNA and is sometimes used as an additional consideration in immunotherapy discussion.

Not a diagnosis and not final medical advice. Must be reviewed by a doctor.
Plain explanation
TMB calculates how many changes are found when the tumor's genetic material is read. The numbers are reported as the number of mutations per unit size.
Clinical meaning
High TMB values in some tumors were associated with greater likelihood of benefit from immunotherapy, but this association was not consistent across cancer types. The thresholds used differ between laboratories and between platforms.
Testing methods
- Next-generation sequencing (NGS)
A test that reads many genes from one tumour sample to find actionable changes.
A negative result does not always mean the marker is absent
Test sensitivity differs by sample type and method. Results need to be read together with the pathology report and clinical condition.
What needs verification
- Platform and panel used, because results cannot be compared directly between platforms.
- The threshold that a laboratory uses to declare high or low.
- Compliance with other markers such as PD-L1 or MSI before interpreting the results.
